History of chromatography. Basic physical-chemical principles of chromatography. Liquid chromatography - individual techniques based on different separation mechanisms, instrumentation, applications, current trends. Gas chromatography - individual techniques based on different separation mechanisms, instrumentation, applications, current trends. Interpretation of chromatographic data, qualitative and quantitative analysis.

Learning outcomes of the course unit

Successful completion of the course the student will gain the following knowledge and skills:
1. Students will be familiarized with the theory of chromatographic separation process and they will be able to apply theoretical knowledge in optimization of chromatographic separation
2. They will be provided with knowledge of instrumentation in liquid, gas and supercritical fluid chromatography
3. They will be able to assess the suitability of chromatographic techniques for solving specific analytical problems and critically evaluate the results.

Prerequisites

Thorough knowledge of physical chemistry, good knowledge of common, inorganic, organic and analytical chemistry.

Planned learning activities and teaching methods

The course uses teaching methods in form of Lecture - 2 teaching hours per week. The e-learning system (LMS Moodle) is available to teachers and students.

Course curriculum

1. Introduction, history of chromatography, classification of chromatographic techniques
2. Basic theory of chromatography – thermodynamical and kinetic aspects, effectivity of chromatographic separation
3. Planar chromatography
4. Column chromatography – basic configuration and theory
5. High performance liquid chromatography – separation principles, instrumentation, columns and rules of their selection, normal and reversed-phase systems, isocratic and gradient elution, ion chromatography, ion-pair chromatography, hydrophobic interaction liquid chromatography, size exclusion chromatography, bioaffinity chromatography, current trends – UPLC, UHPLC
6. Gas chromatography – separation principles, instrumentation, capillary and packed columns, rules for their selection, classification of stationary phases in gas chromatography, derivatization in gas chromatography
7. Tandem techniques in chromatography – systems LC-LC, LC-GC, GC-GC, GCxGC
8. Qualitative aspects of chromatographic data – correlation structure-retention, retention indices in gas chromatography
9. Quantitation in chromatography

Aims

The aim of this course is:
- Thorough familiarization of students with chromatography as an analytical separation technique
- detailed discussion of chromatographic separation mechanisms, including their capabilities and limitations
- introduction of students to the field of chromatography application in analytical practice

Specification of controlled education, way of implementation and compensation for absences

Attendance at lectures is optional and is not controlled, but it is recommended.
