Original Abstract

DNA and RNA are compounds, very familiar to us. They are key to understanding of living matrix. All important processes, including the most fascinating such as the coding the amino-acids, are accompanied by charge transfers between each nucleotide. The range of the charge can be recorded on the basis of various measurements, for example the fluorescence, which depends on many conditions. In this case, we used steady state fluorescence to determine the emission and excitation spectras and Time Correlated Single Photon Counting method to determine lifetime of the fluorescence. Four samples of synthetized oligomers of twelve nucleotides with 2-aminopurine as electron acceptor were analyzed. To characterize the influence of temperature on charge transfer, redox-active samples of DNA-DNA and RNA-DNA were compared with reference redox-inactive counterparts. The redox-active oligomers have guanine as electron donor separated by two A-T or A-U basepairs. From the excitation spectras and differences in the 270-280nm region we describe the influence of temperature on the extent of base stacking of 2-aminopurine to the chain. From the decrease of the redox active oligomers fluorescence relative to the redox-inactive counterparts, the extent of charge transfer is determined and related to the temperature. The results of these measurements might positively contribute to the further research in the field of biosensoric medicine and for better and more precise understanding of human body processes.
