Original Abstract

One of the recent trends in the field of scanning electron microscopy deals with observation of soft biological specimens at higher pressure in the specimen chamber. This microscopy is called environmental or low vacuum or variable pressure scanning electron microscopy. As wet biological specimens are to be observed in their natural state, specific process of hydration and temperature conditions in the dependence on pressure in the specimen chamber must be ensured. Two methods for observation of wet biological specimens in their unchanged or only a slightly changed natural state using the environmental scanning electron microscope are described in this article.
