Original Abstract

This work was focused on monitoring of covalent cisplatin bonding with lambda DNA fragment. The PCR fragment with a length of 498 bp and 100 uM cisplatin infusion solution was used. Interactions of DNA fragment were observed electrophoreticallyand spectrophotometrically. Experiments show, that during denaturation, cisplatin significantly reduces the melting temperature of DNA fragment. The average melting temperature was determined to 71 degrees C. After addition of cisplatin (100 uM), temperaturedecreased by 15 degrees C on average.
