Projekt studuje fyzikálně-chemické interakce hyaluronan-tenzid a hyaluronan-aminokyselina ve vodném prostředí. Soustředí se zejména na biokompatibilní tenzidy a aminokyseliny s povrchově aktivními vlastnostmi. Účelem projektu je nalézt v uvedených systémech asociativní koloidy schopné solubilizovat hydrofobní látky, které by byly vhodnými kandidáty pro cílenou distribuci nepolárních aktivních látek, zejména léčiv. Dále projekt rozvine aplikaci ultrazvukové spektroskopie s vysokým rozlišením v oblasti studia interakcí biopolymer-tenzid.

Project is focused on study of physico-chemical interactions in hyaluronan-surfactant and hyaluronan-amino acid systems. Interactions in these systems should result in formation of self-assembling structures that can be used in the targeted delivery of hydrophobic active substances, drugs, in particular. Hydrophilic, highly hydrated hyaluronan chain which has a specific biological function, which is recognized and bound by cells through specific receptors plays the role of transporting and targeting agent. Surfactants or amino acids should provide non-polar domains solubilizing the hydrophobic substance. In particular, biocompatible surfactants like polar lipids or acylglycosides will be investigated. Simple amino acids will be used to obtain basic information on hyaluronan-amino acid interactions whereas alkylated (acylated) amino acids and betaine surfactants will be used in the development of the carrier systems. Physico-chemical interactions between hyaluronan chain and surfactant/amino acid aggregates (micelles), containing the solubilized hydrophobe, will form the final delivery system, which, at the target, will decompose to its biodegradable constituents. Study on influence of temperature, pH and ionic strength is included. Further, the project will develop applications of the high resolution ultrasound spectroscopy in research of biopolymer-surfactant interactions.
