Laboratory Classes in Special Toxicology

Practical course of special toxicology to increased knowledge of special toxicology and will allow connection of theoretical and practical knowledge. The scope of practical course is to introduce students with the possibilities of determination of some toxic substances in biotic and abiotic matrices and determination of selected toxicological data directly from an experiment in laboratory conditions. Students will gain an overview of selected experimental methods used in toxicology, preparation of abiotic and biotic matrices for the determination of selected toxic compounds. Students will be introduced to a screening technique TLS (1D, 2D), the instrumental technique used in toxicology (atomic absorption spectrometry - GF AAS, F AAS), methodology and evaluation of toxicity tests (test viability, cytotoxicity test). The analysis and evaluation of case studies from toxicology are included of practicum of special toxicology.

Learning outcomes of the course unit

Students will acquire new skills, knowledge and experience in these areas and increase the competence of students as follows:

1st Students will gain an overview of selected experimental methods used in toxicology.
2d Students will be able to prepare of abiotic and biotic matrices for the determination of selected toxic compounds.
3rd Students will know a screening technique TLS (1D, 2D).
4th Students will know instrumental technique used in toxicology (atomic absorption spectrometry).
5th Students will know the methodology and evaluation of toxicity tests (Test with neutral red - test viability, MTT test - cytotoxicity tests, trypan blue exclusion test - a test of viability and cytotoxicity).
6th Students will be able will be able to evaluate the case studies.

Prerequisites

Knowledge of general toxicology, from the lectures of special toxicology of analytical chemistry and environmental chemistry,Practicum of Analytical chemistry I and II.

Planned learning activities and teaching methods

The course uses teaching methods in form of Laboratory exercise - 2 teaching hours per week. The e-learning system (LMS Moodle) is available to teachers and students.

Assesment methods and criteria linked to learning outcomes

Evaluation includes preparation of a project and its presentation on a selected topic from the list of Case studies and all required protocols from the practical courses.

1st Introduction to practice, work safety in toxicology laboratory
2nd Extraction procedures for the preparation of biotic and abiotic matrices
3rd Identification of the type of nicotine alkaloids in Nicotiana tabacum
4th Identification of biologically active compounds (derivatives Pyranocumarin)
6th Application of TLC of determine PAHs and separation compounds with using 2D TLC
7th Identification and quantification of toxic metals toxic metals (cadmium, arsenic) in abiotic and biotic matrices with using GF AAS
8th Identification and quantification of toxic metals toxic metals (chromium, lead) in biotic and abiotic matrices with using F AAS
9th Test with neutral red - test viability
10th MTT test - cytotoxicity tests
11th Test with trypane blue - test viability and cytotoxicity
12th Case Study of toxicology

Aims

The aim of this course is to prepare abiotic and biotic matrices for the determination of selected toxic compounds, screening techniques TLC (1D, 2D), instrumental techniques used in toxicology (atomic absorption spectrometry). Other aim of this course is to introduce students to the problems of toxicity tests and students will learn the methodology and evaluation of toxicity tests (test viability, MTT test - cytotoxicity test, test viability and cytotoxicity). Next goal of the course is evaluation of case studies of toxicology.

Specification of controlled education, way of implementation and compensation for absences

Practical course is supported by the eLearning courses, in which are available support materials for students, including electronic textbooks and other supplementary materials. In the second half of the semester the consultations are reserved under which it is possible to consult processing protocols. Submission of satisfactory protocols from each practical training and participation of at practices is required (minimum is 100%). Students of combined study programs will complete a practical course under the block schedule.
